Boston Fern Culture

MEDIA: Prepare MS medium with PPM and no hormones as described in the manual.

    ISOLATION AND CULTURE

  • Collect clean 2-4 cm (0.75 - 1.5 inches) fern runners and discard all but the terminal 2 inches.
  • Dip the runners in 70% alcohol for a few seconds.
  • Place in beaker containing 10% commercial bleach (1/4 cup bleach + 2 1/4 cup water + few drops of dish detergent) and soak 7 minutes. Stir occasionally  using a long forceps or spatula.
  • Transfer explants to a jar containing sterile water and soak for 2-3 minutes to rinse off  the bleach.
  • Place one runner tip on a sterile plate (wipe off with 70% alcohol).
  • Remove the terminal  1/2 inch with a sterile knife (that had been dipped in 70% alcohol).
  • Place in a baby food jar containing MS medium with no hormones.  The plant piece can be laid horizontally on the meduim surface or stuck in the medium ("up and down").
  • Replace cap, wrap/seal with florists' tape and incubate on a lighted shelf or in a well lite room out of direct sun light.
