African Violet Leaf
Tissue Culture

Plant tissue culture involves the growth of a plant cell, tissue, or organ in a sterile environment. The plant piece is disinfected and placed in an environment which will induce growth of axillary buds or adventitious shoots (shoots that develop from places other than a bud). Hundreds of plants can be developed from a single piece of leaf. See the photos on the next two pages.

 

Prepare MS medium with 1 ml PPM and BAP as described in the manual.

Prepare the "clean box" as described in the manual. Spray or wipe culture bottles and salad plate with 70% alcohol.

In the "clean box" sterilize forceps and knife by dipping in 70% alcohol. Dip each leaf in 70% alcohol for about 1 minute.

 

Place leaf in 10% bleach solution (1/4 cup bleach + 2 1/4 cup water + few drops of dish detergent) and soak for 10 minutes. Stir occasionally using the forceps. Also use the forceps to hold floating leaves below the surface of the solution.

After 10 minutes, place bottle with bleach + leaves in the clean area. Remove one leaf at a time and place in jar of sterile water. Soak  leaves in the sterile water until you are ready to cut and culture them. 
 

 

In the clean area, transfer leaves to a sterile plate.

 

Cut away outside edges of leaf using the sterile knife and the sterile forceps. Then cut the remaining leaf into 2-4 sections. Place one leaf section into each baby food jar containing African Violet Growth Medium. Press each piece slightly into the medium. Replace cover and seal with florists' tape.
